Absence of Nitrergic Modulation of Starling Response in Haemoglobin-Less Antarctic Fish Chionodraco hamatus

چکیده

The Frank-Starling response is an intrinsic heart property that particularly evident in the fish because piscine cardiomyocytes are extremely sensitive to stretch. Several mechanisms and compounds influence response, including free radical nitric oxide produced by synthases vascular endothelium of all vertebrates. Besides its role scavenging oxide, hemoglobin may act as a source transporter. In this context, hemoglobin-less Antarctic teleost Chionodraco hamatus (icefish) represents unique opportunity investigate involvement response. Using isolated perfused preparation, weverified basal nitrergic tone not implicated addition, comparing expression activation C. red-blooded Trematomus bernacchii, we found endothelial isoform synthase (the primary generator during shear stress) be less expressed activated former.

Laser microdissection-based analysis of the Y sex chromosome of the Antarctic fish Chionodraco hamatus (Notothenioidei, Channichthyidae)

Microdissection, DOP-PCR amplification and microcloning were used to study the large Y chromosome of Chionodracohamatus, an Antarctic fish belonging to the Notothenioidei, the dominant component of the Southern Ocean fauna. The species has evolved a multiple sex chromosome system with digametic males showing an X1YX2 karyotype and females an X1X1X2X2 karyotype. Fluorescence in situ hybridizatio...

Characterisation of intestinal peptide transporter of the Antarctic haemoglobinless teleost Chionodraco hamatus.

H(+)/peptide cotransport was studied in brush-border membrane vesicles (BBMV) from the intestine of the haemoglobinless Antarctic teleost Chionodraco hamatus by monitoring peptide-dependent intravesicular acidification with the pH-sensitive dye Acridine Orange. Diethylpyrocarbonate-inhibited intravesicular acidification was specifically achieved in the presence of extravesicular glycyl-L-prolin...

A New APEH Cluster with Antioxidant Functions in the Antarctic Hemoglobinless Icefish Chionodraco hamatus

Acylpeptide hydrolase (APEH) is a ubiquitous cytosolic protease that plays an important role in the detoxification of oxidised proteins. In this work, to further explore the physiological role of this enzyme, two apeh cDNAs were isolated from the Chionodraco hamatus icefish, which lives in the highly oxygenated Antarctic marine environment. The encoded proteins (APEH-1(Ch) and APEH-2(Ch)) were ...

Phospholamban S-nitrosylation modulates Starling response in fish heart.

The Frank-Starling mechanism is a fundamental property of the vertebrate heart, which allows the myocardium to respond to increased filling pressure with a more vigorous contraction of its lengthened fibres. In mammals, myocardial stretch increases cardiac nitric oxide (NO) release from both vascular endothelium and cardiomyocytes. This facilitates myocardial relaxation and ventricular diastoli...

H+/peptide cotransport was studied in brush-border membrane vesicles (BBMV) from the intestine of the haemoglobinless Antarctic teleost Chionodraco hamatus by monitoring peptide-dependent intravesicular acidification

provided by dietary protein digestion is mediated by electrogenic brush-border membrane (BBM) transport processes. Peptide influx, coupled to an inwardly directed H+ gradient, is additionally driven by the inside-negative transmembrane electrical potential (Ganapathy et al., 1994). Peptide transporters are unique among the solute transporters in presenting H+ as the cotransported species and th...
